| Text of abstract: | The survival of projection neurons and various types of interneurons in the mouse striatum after transient focal cerebral ischemia were studied using immunocytochemical and histochemical techniques and stereological procedures. At one week after 30 min of middle cerebral artery occlusion, various striatal neurons exhibited differential susceptibility to ischemic damage. The order of vulnerability was: projection neurons > parvalbumin-expressing interneurons > nitric oxide synthase-containing interneurons > cholinergic interneurons. Within the ischemic lesion, projection neurons were almost completely lost whereas cholinergic interneurons were spared one week after the ischemic insult. The present study shows the usefulness of the intraluminal filament model for inducing transient focal ischemia in mice, allowing for long-term survival of the animals and, combined with stereology, unbiased quantification of various neuronal populations. The differential vulnerability of various striatal neurons to ischemic damage may be due, e.g., to differences in glutamate receptor composition, trophic support, and expression of antiapoptotic proteins. |
